چکیده

Single molecule localization based super-resolution imaging techniques require repeated localization of many single emitters. We describe a method that uses the maximum likelihood estimator to localize multiple emitters simultaneously within a single, two-dimensional fitting sub-region, yielding an order of magnitude improvement in the tolerance of the analysis routine with regards to the single-frame active emitter density. Multiple-emitter fitting enables the overall performance of single-molecule super-resolution to be improved in one or more of several metrics that result in higher single-frame density of localized active emitters. For speed, the algorithm is implemented on Graphics Processing Unit (GPU) architecture, resulting in analysis times on the order of minutes. We show the performance of multiple emitter fitting as a function of the single-frame active emitter density. We describe the details of the algorithm that allow robust fitting, the details of the GPU implementation, and the other imaging processing steps required for the analysis of data sets.

Precise multi-emitter localization method for fast super-resolution imaging.

We present a method that can simultaneously locate positions of overlapped multi-emitters at the theoretical-limit precision. We derive a set of simple equations whose solutions give the maximum likelihood estimator of multi-emitter positions. We compare the performance of our simultaneous localization analysis with the conventional single-molecule analysis for simulated images and show that ou...

Mapping molecules in scanning far-field fluorescence nanoscopy

In fluorescence microscopy, the distribution of the emitting molecule number in space is usually obtained by dividing the measured fluorescence by that of a single emitter. However, the brightness of individual emitters may vary strongly in the sample or be inaccessible. Moreover, with increasing (super-) resolution, fewer molecules are found per pixel, making this approach unreliable. Here we ...

Shifting molecular localization by plasmonic coupling in a single-molecule mirage

Over the last decade, two fields have dominated the attention of sub-diffraction photonics research: plasmonics and fluorescence nanoscopy. Nanoscopy based on single-molecule localization offers a practical way to explore plasmonic interactions with nanometre resolution. However, this seemingly straightforward technique may retrieve false positional information. Here, we make use of the DNA ori...

The lateral and axial localization uncertainty in super-resolution light microscopy.

A study of the uncertainty of localizing single-molecule emitters for super-resolution light microscopy is presented. Maximum likelihood estimation (MLE) is found to be superior to least-squares fitting for low background levels, but the performance difference between the two methods decreases to a few percent for practical background levels. It is shown that the performance limit of MLE, the C...

Fast and Precise 3D Fluorophore Localization based on Gradient Fitting

Astigmatism imaging approach has been widely used to encode the fluorophore's 3D position in single-particle tracking and super-resolution localization microscopy. Here, we present a new high-speed localization algorithm based on gradient fitting to precisely decode the 3D subpixel position of the fluorophore. This algebraic algorithm determines the center of the fluorescent emitter by finding ...
